Clarifying the cardiac proteasome paradox: protein quality control.
نویسنده
چکیده
Correct folding of proteins is crucial for proper protein, cell, and organ function.1 Mutations and cellular stresses, some of which occur during pathology, can lead to incorrect folding and subsequent loss of protein function. If it goes uncorrected, then this loss of protein function, along with the toxicity associated with the accumulation of misfolded proteins in cells, results in eventual organ failure.2 Many diseases, including neurodegenerative, hepatic, endocrine, and cardiovascular disorders, are thought to be associated with, if not caused by, the organ failure resulting from the accumulation of misfolded proteins.3 For example, in the heart, accumulation of misfolded proteins has been associated with hypertrophic and dilated cardiomyopathies as well as ischemic heart disease.4

Proteasome functional insufficiency in cardiac pathogenesis.
The ubiquitin-proteasome system (UPS) is responsible for the degradation of most cellular proteins. Alterations in cardiac UPS, including changes in the degradation of regulatory proteins and proteasome functional insufficiency, are observed in many forms of heart disease and have been shown to play an important role in cardiac pathogenesis. In the past several years, remarkable progress in und...
متن کاملThe effect of a period of resistance training on the muscle proteasome activity and autophgy flux in mice with cancer induce cachexia
Introdution: Muscle weakness that is caused by cancer called Cachexia. One of the causes of the formation of the cachexia is the change in protein degradation, and the ubiquitous protease and Autophagy Lysosomes system is the most important protein breakdown system. Resistance training has been one the best stimulator of increasing muscular mass. Therefore, the purpose of this study was to inve...
متن کاملCytokine-induced oxidative stress in cardiac inflammation and heart failure—how the ubiquitin proteasome system targets this vicious cycle
The ubiquitin proteasome system (UPS) is critical for the regulation of many intracellular processes necessary for cell function and survival. The absolute requirement of the UPS for the maintenance of protein homeostasis and thereby for the regulation of protein quality control is reflected by the fact that deviation of proteasome function from the norm was reported in cardiovascular pathologi...
متن کاملThe bitter end: the ubiquitin-proteasome system and cardiac dysfunction.
Many elements contribute to congestive heart failure: changes in perfusion, hemodynamic stresses, alterations in calcium metabolism, and dysregulation of cell signaling pathways. Intervention in these processes forms the basis for current heart failure therapies. Nevertheless, heart failure is primarily a disease of wear and tear; despite everything we know about cardiac physiology and the clin...
متن کاملRole of the ubiquitin proteasome system in the heart.
Proper protein turnover is required for cardiac homeostasis and, accordingly, impaired proteasomal function appears to contribute to heart disease. Specific proteasomal degradation mechanisms underlying cardiovascular biology and disease have been identified, and such cellular pathways have been proposed to be targets of clinical relevance. This review summarizes the latest literature regarding...
